The problem
Methanol induction (AOX1 promoter) is the Pichia standard, but industrial settings increasingly require methanol-free processes for safety and scale-up reasons. Alternative promoters (pGAP, pFLD1, pPET) have different expression characteristics, and the optimal construct design may differ — but most design tools default to AOX1 assumptions.
